SLOPP-Y is a restriction enzyme that cuts anywhere it encounters a CA sequence in between the C and the A.

Draw a line anywhere the restriction enzyme SLOPP-Y will cut! Remember to read the bottom strand from right to left.

D7S280 is an STR found on human chromosome 7 that repeats the sequence GATA. The DNA sequence of the representative allele of this locus is shown. Find the repeat sequence GATA. How many repeats are shown on the DNA sequence? Different alleles of this locus may have from 6 to 15 tandem repeats of GATA.

One of the hotel guests that was not accounted for after the fire was a tourist, John Doe, visiting Reno from New York. Authorities sampled DNA from his toothbrush obtained from his apartment in New York. They used the DNA to generate a DNA profile using the 13 standard STR loci. The data is shown in Figure 2.

The population of the United States in 2006 was about 298.4 million (2.98 × 108). Based on this information, explain why the medical examiner was confident in telling the victim’s family that the recovered remains were those of their son.

List 2 precautions the officer is taking to prevent contamination.

Background

Anna Anderson claimed for more than 60 years that she was Princess Anastasia, the Grand Duchess of Russia, daughter of Tsar Nicholas and Tsarina Alexandra. Anna Anderson knew a great deal about life in the Russian palace and she could speak very knowledgably about family members. Anna had many people convinced that she was Anastasia, but does the DNA evidence support her claim?

In this activity, you will examine the STR profiles of Anna Anderson, Tsar Nicholas, and Tsarina Alexandra to determine if the DNA evidence supports the claim that Anna Anderson was the child of the tsar and tsarina. Recall that each parent must contribute one STR allele for each STR marker.
